Pogostemonis Herba is well known to be used as Chinese herbal medicine to remove dampness,relieve sunstroke,stop vomiting and stimulate the appetite.Patchouli alcohol is the most active ingredient in the essential oil of Pogostemonis Herba(Patchouli oil).In this study,a simple and selective GC/MS method was developed and validated for the determination of patchouli alcohol in rat plasma.The method was validated in terms of selectivity,linearity and sensitivity,and showed advantages in monitoring the pharmacokinetic behaviors of patchouli alcohol following single-dose oral administration in rats.The influence of some other components in patchouli oil on the pharmacokinetic profile of patchouli alcohol was investigated as well.Results revealed the pharmacokinetic behaviors were linear dynamics both in patchouli alcohol and patchouli oil groups.The t1/2z was significantly greater in the 20-mg dose(35.10 [13.09] h)compared with the 6-mg(17.83 [7.10] h; P =0.015)and the 2-mg(15.28 [3.68] h; P =0.014)after patchouli oil administration.The Cmax of patchouli alcohol was greater with three doses in patchouli alcohol group compared with patchouli oil group.Meanwhile,the Tmmax was prolonged in oil group.The components in patchouli oil had influence on the pharmacokinetic behaviors of patchouli alcohol in the process of absorption and elimination.This is the first report on the determination of patchouli alcohol in rat plasma after oral administration.These results provide a meaningful basis for evaluating the clinical application of this medicine.
