A systems biology approach was developed to study in detail the effect of HER2 inhibitors (trastuzumab, pcrtuzumab) on the response of the ERK/PI3K/PTEN/AKT signalling network, one of dominant signalling pathways in cancer progression, the source of drug resistance, and a promising target of cancer therapy.The integrated systems framework developed includes iterated in silico, in viro experiments, and analysis in clinical cohorts, allowing cyclic generation and validation of hypotheses for mechanisms of signalling robustness.
