Objective: Squamous cell carcinoma (SCC), such as gingival carcinoma, has been recognized to display a tendency of bone destruction at the late stage of the disease.However, the mechanism involved in this process is still not clearly elucidated.The study was to investigate whether bone microenvironment could alter the histological characteristics of the invaded SCC cells.Methods: Human tongue carcinoma cell line Tca83 cells and cervical carcinoma cell line HeLa cells (3× 106cells/mouse) were respectively injected into the erector spinae of the 6-week-old female BALB/c nude mice to create the bone destruction model caused by SCC cells.At 30 days post-tumor cell inoculation, the mice were scarified and the tumor with its invaded spine was removed, immediately fixed by 10% formalin, decalcified in 17% EDTA solution, embedded in paraffin wax, sectioned into 4-μm slices, then subjected to hematoxylin and eosin (HE) and immunostaing of IL-6, IL-11, TNF-a, MMP-2, MMP-3, MMP-9, MMP-13, Runx-2, BSP and OCN.
