纳米通道单分子检测技术是一种根据单个生物分子在电场力驱动下穿过纳米尺寸孔道时产生的特征离子流信号在单个分子水平上研究生物分子个体行为的分析方法.探索将单个Stable Protein 1(SP1)、α-Hemolysin(α-HL)生物纳米通道作为"弱力"识别传感器,实时监测了帕金森致病蛋白单个α-synuclein单体从伸展状态形成部分折叠的中间体以及实现分析肿瘤抑制蛋白P53与双链DNA之间的"弱"相互作用.通过合成系列多肽-DNA conjugations,在单分子水平上获得反映多肽-DNA conjugations通过单个α-HL纳米通道的直接电信号特征谱图.
