Production of maternal haploids via intra-specific genotypes as the haploid inducer is routine and highly efficient in maize.However,the underlying mechanism of haploid induction (HI) is unclear.Triphenyltetrazolium chloride (TTC) staining data indicated that the viability,rather than fertility,of the inducers mature pollen is lower than that of regular lines.We have developed a method to isolate the three nuclei from a mature pollen grain and four microspores from a tetrad for whole-genome sequencing.
