By screening of expressed genes, the chalcone synthase (CHS) gene was obtained from the wheat (Triticum aestivum L), infected by plant pathogen Gaeumannomyces graminis var.tritici.The full-length DNA of the CHS gene was obtained from genomic DNA of Triticum aestivum L , named TaCHS.Sequence analysis indicated that TaCHS was 2543 bp in full length and contained a promoter region of 1264 bp, an intron of 94 bp, and an opening reading frame (ORF) of 1185 bp encoding a 394 predicted amino acids residues which possessed all the conserved active sites for the CHS function as well as the family signature.Sequence alignment and phylogenetic analysis of the deduced amino acids revealed that TaCHS shared more than 88% of homology with CHS from other reported plants in Gramineae.The TaCHS gene from Triticum aestivum was closely related to the gene from Secale cereal, Thinopyrum ponticum and Triticum urartu.The promoter region of TaCHS gene contained fungal elicitor responsive elements.It also had such typical cis-acting elements as including light responsive elements, phytohormone responsive elements, Myc and Myb binding site, TATA-Box and CAAT-Box.
