紧密连接蛋白主要位于上皮细胞近管腔的侧面，其功能是封闭细胞间隙，阻止管腔上皮层内外物质的自由进出，是上皮细胞选择通透作用的物质基础.紧密连接为多种蛋白相互作用下形成的复合结构，主要由跨膜蛋白和胞质蛋白两种成分组成，其中主要跨膜蛋白有occludin和claudin-1.
