Methylcobalamin (methyl-vitamin B12), a vitamin B12 analog, is used in the treatment of peripheral neuropathy and diabetic neuropathy.It is also applied as a preliminary treatment for amyotrophic lateral sclerosis.In some clinical trials, it has been observed that methylcobalamin can relieve neuropathy pain symptoms as an auxiliary chemical medicine.However, the effect of Methylcobalamin on pain signals remains unclear.The aim of this study was to determine whether methylcobalamin inhibits the spontaneous discharge from primary afferents in the periphery.In CCD (a chronic compression of DRG) model of rats,single fiber recording technique was used in the present study.Methylcobalamin exhibited an inhibition of spontaneous activity from A-type DRG neurons in CCD rats in a dose-dependent manner from 3 to 300 μmol/L.The inhibition of methylcobalamin in A-type CCD neurons was shown to be reversible in most cases (30/36) within 10 mins after washout.The inhibitory percentage was 34±6.4%(p<0.05, n=6), 56±6.9%(p<0.05, n=6), 94±1.8%(p<0.05, n=6) at the concentration of 30 μmol/L, 100 μmol/L and 300 μmol/L respectively.The 50% inhibitory concentration of methylcobalamin is 76 μmol/L.In the present study it was showed an inhibitory effect of methylcobalamin on the spontaneous activity from A-type DRG neurons.It is suggested that methylcobalamin may serve as a candidate of peripheral analgesic drugs in neuropathic pain state.
