Folic acid mediated solid lipid nanocarriers loaded with docetaxel and oxidised single-walled carbon

  Single-walled Carbon nanotubes (SWNT) has large near-infrared absorption coefficient, large surface area and has shown great potential in drug delivery.In this study, we obtained ultra short OSWNT using mixed acid oxidation method, then conjugated DTX and folic acid with OSWNT via π-π accumulation or through ester linkage or amide linkage.A targeting and photothermal sensitive drug delivery system FA-DTX-OSWNT-SLN was prepared following a microemulsion technique.The size and zeta potential of FA-DTX-OSWNT-SLN were 182.8±2.8nm and-34.59±1.50mV, respectively.TEM images indicated that that FA-DTX-OSWNT-SLN was spherical and much darker than general SLN maybe because of the exit of OSWNT, OSWNT winds round or inserts into or was encapsulated into the nanocarriers.Compared with free DTX, FA-DTX-OSWNT-SLN could efficiently cross cell membranes, lead to more apoptosis and afford higher antitumor efficacy in a cultured MCF-7 cells in vitro.Meanwhile, the combination of 808nm near-infrared laser irradiation significantly enhanced the cells inhibition.In conclusion, FA-DTX-OSWNT-SLN drug delivery system combination with 808nm NIR laser irradiation may be promising for targeting and photothermal cancer treatment with multiple mechanisms in future.
