蚯蚓蛋白酶HBeAgases胃肠吸收的研究

  HBeAgases 是一组蚯蚓蛋白酶,具有降解纤连蛋白和乙型肝炎e 抗原的作用。免疫组织化学的结果显示,HBeAgases 由蚯蚓的上消化道产生,定位于环节之上的消化道系统内。为了研究HBeAgases 在肠道内的吸收,本文作者研究了这些蛋白酶在肠壁和血液内的分布。采用HBeAgases 喂食小鼠,通过EFE-Ⅲ-1 的抗体进行标记可以观察到,HBeAgases 分布在胃肠壁的囊泡内,蛋白酶的含量分别是十二指肠最高,其次是小肠,回肠壁内有零星分布,但大肠壁内几乎没有分布。采用Caco2 细胞建立单细胞层作为模型,同样证明了HBeAgases 能够通过细胞。采用ELISA 测定结果显示,蚯蚓蛋白酶在血清内的含量20 分钟开始出现,60分钟到达峰值,之后含量逐渐降低。这些结果提示,蚯蚓蛋白酶可能以囊泡的形式主要在十二指肠内通过肠道壁吸收入血。阐明HBeAgases 通过肠壁吸收的机制,不但对生物大分子直接吸收的研究具有理论意义,同时也有助于蚯蚓蛋白酶在治疗乙型病毒性肝炎应用方面具有潜在的使用价值。
