The method was proposed to predict retention times and peak shapes simultaneously in gradient elution under reversed-phase high-performance liquid chromatography (RP-HPLC) based on the isocratic elutioned retention datas using the plate theory. The method can give good prediction result with different retention equations of three or four-parameter when tested in the retention prediction by 16 common sulfonamides(SAs) in 18 kinds of gradient conditions. In addition, the method of using the plate theory can well predict the change of peak shapes where caused the expansion or compression of peak under different gradient conditions.
