露湿漆斑菌(Myrothecium roridum)是一种温带和热带普遍存在的土壤寄生菌,可侵染大豆、棉花、扁豆、茄、辣椒、甜菜、番茄、苘麻、蓖麻、锦葵等寄生18科34种作物经济作物,该真菌主要通过产生单端孢霉烯毒素引起经济作物产生严重的叶斑、疫病以及果实腐烂等症状,对大豆、棉花和绿豆等经济作物的产量造成了严重的损失,而且露湿漆斑菌单端孢霉烯毒素还会通过这些经济作物进入人畜体内,严重威胁人畜健康.

露湿漆斑菌(Myrothecium roridum)是一种温带和热带普遍存在的土壤寄生菌,可侵染大豆、棉花、扁豆、茄、辣椒、甜菜、番茄、苘麻、蓖麻、锦葵等寄生18科34种作物经济作物,该真菌主要通过产生单端孢霉烯毒素引起经济作物产生严重的叶斑、疫病以及果实腐烂等症状,对大豆、棉花和绿豆等经济作物的产量造成了严重的损失,而且露湿漆斑菌单端孢霉烯毒素还会通过这些经济作物进入人畜体内,严重威胁人畜健康.本课题组前期在广藿香内生真菌露湿漆斑菌种分离得到了三种不同结构的单端孢霉烯毒素,表现出了极强的细胞毒性.因此,本课题组采用Illumina Hiseq 2500测序平台对30℃培养3天、5天、7天和9天的露湿漆斑菌混合样品进行了de novo转录组测序,获得了完整的露湿漆斑菌转录组序列.测序结果获得9.00Gb Clean Data,Q30碱基百分比为85.07%.De novo组装后共获得35,485条Transcript, 25,996条Unigenes.其中长度在lkb以上的Unigene有8,576条.对Unigene进行功能注释,包括与NR、Swiss-Prot、KEGG、COG、GO数据库的比对,共获得18,199条Unigene的注释结果.注释获得了包括单端孢霉烯合成酶在内的10多种单端孢霉烯生物合成相关基因的序列,通过blastx将Unigene序列比对至蛋白数据库,对蛋白功能进行注释并进行分类统计,从宏观上了解露湿漆斑菌的基因功能分布特征.经转录组测序分析比对,在露湿漆斑菌中预测到单端孢霉烯生物合成相关的基因Tri3、Tri4、Tri5、Tri6、Tri8、Tri12和Tri15等,为后期阐明露湿漆斑菌的致病机制并控制露湿漆斑菌对经济作物的危害奠定理论基础.
