目的探讨壳寡糖诱导肿瘤细胞凋亡机制,为壳寡糖开发应用提供实验依据。方法选择小鼠体内注射的方法将壳寡糖注入荷瘤小鼠体内,观察肿瘤生长情况,计算抑瘤率,流式细胞仪测定细胞凋亡率,电镜观察肿瘤细胞超微结构。原位杂交法检测Survivin mRNA表达。结果壳寡糖抑瘤率为42.03%,流式细胞仪检测壳寡糖凋亡率20.68%,与模型组比较,差异有统计学意义。壳寡糖组镜下可见瘤细胞以凋亡变化为主,壳寡糖下调肿瘤细胞SurvivinmRNA表达,与模型组比较,差异具有统计学意义。结论壳寡糖可诱导肿瘤细胞凋亡,其机制可能与下调Survivin mRNA表达密切相关。
OBJECTIVE To investigate the mechanism of chitooligosaccharides inducing apoptosis of tumor cells and provide experimental evidence for the development and application of chitooligosaccharides. Methods Chitosan oligosaccharides were injected into the tumor-bearing mice by injection in vivo. The tumor growth was observed, the tumor inhibition rate was calculated, the apoptosis rate was measured by flow cytometry, and the ultrastructure of tumor cells were observed by electron microscope. Survivin mRNA expression was detected by in situ hybridization. Results The inhibitory rate of chitooligosaccharides was 42.03%. The apoptosis rate of chitooligosaccharides by flow cytometry was 20.68%. Compared with the model group, the difference was statistically significant. Chitooligosaccharides microscopy showed tumor cells to apoptosis mainly chitooligosaccharides down regulation of tumor cells SurvivinmRNA expression, compared with the model group, the difference was statistically significant. Conclusion Chitooligosaccharides can induce tumor cell apoptosis, and its mechanism may be related to down-regulation of Survivin mRNA expression.
