在高等真核生物中,CTCF是一种结构高度保守、功能多样,拥有十一个锌指DNA结合结构域的染色质绝缘子结合蛋白.CTCF在基因活化与沉默、绝缘功能、基因组印记、X-染色体失活以及基因组核内空间组织等不同生理过程中发挥关键作用.我们发现RNA解螺旋酶p68和非编码RNA SRA在基因组水平上参与调控凝集蛋白复合物和绝缘子结合蛋白CTCF的转录绝缘功能;p68和SRA具有调控CTCF介导的增强子与远距离H19/Igf2启动子之间相互作用的能力,从而调节印记基因H19和Igf2的表达模式;最近,我们进一步发现降低CTCF水平能够抑制癌细胞中核糖体RNA前体的转录,并减缓癌细胞增殖,同时,发现Condensin复合物负向调控CTCF介导的核糖体rRNA基因的转录是RNA聚合酶I依赖型的;通过运用RIP-seq的技术手段,我们进一步寻找与CTCF相互作用的特异性LncRNA(s),目前正在对部分LncRNA(s)的功能进行进一步研究.
