The lineage specific basic helix-loop-helix transcription factor SCL (Stem Cell leukemia) is critical for the formation of hematopoietic lineages during normal hematopoiesis and is aberrantly activated in up to 60 % ofpatients with T-cell acute lymphoblastic leukemia (T-ALL).To understand the mechanisms underlying the control of SCL function, we have purified SCL-containing multiprotein complexes and identified the interacting polypeptides by mass spectrometry.We found that SCL forms a complex with histone demethylase LSD1, corepressor CoREST, and HDAC 1/2, which exhibits histone demethylase and deacetylase activities.Furthermore, L SD 1 enhances SCLdependenttranscriptional repression, and the demethylase activity of LSD1 is essential for the repressive activity.
