Alcohol-related myopathy is characterized by severe biochemical and structural changes in skeletal muscle,and muscle at-rophy is one of the main clinical manifestations.Ethanol produces large amounts of reactive oxygen species (ROS) and free radicals in the bodys metabolism,causing oxidative stress and lipid peroxidation in cells.Ubiquitin ligase is a key enzyme in skeletal muscle atrophy.Atrogin-1,a novel E3 ubiquitin ligase,is thought to be the early biomarker of muscle atrophy.In this study,we established the model of acute alcoholism by exposing C2C12 myoblasts to alcohol.C2C12 myoblasts were firstly cultivated with growth medium (GM;10% fetal bovine serum in DMEM) in a CO2 incubator at 37℃ for 2 or 3 days.
