Pancreatic cancer is the fourth leading cause of cancer-related death and the mortality rate remains very high.Despite efforts in the past 50 years, conventional treatment approaches, such as surgery, radiation, chemotherapy, or combinations of these,have had little impact on the course of this aggressive neoplasm.Pancreatic cancer still continues to be a major unsolved health problem at the start of the 21st century.Therefore, to effectively diagnose, prevent, and treat this disease, only by developing a detailed understanding of the molecular biology underlying pancreatic cancer formation and progression will be in a position.CD147, also named extracelluar matrix metalloproteinase inducer (EMMPRIN),is a member of the immunoglobulin family and a glycoprotein enriched on the surface of tumor cells, promotes invasion, metastasis,growth and survival of malignant cells, and confers resistance to some chemotherapeutic drugs.To determine the possible role of CD147 in the invasiveness properties of laryngeal carcinoma, we used RNA interference approach to silence CD147 expression in Panc-i cell line at high level of CD147 expression.We observed that CD147 expression was significantly impeded at both mRNA and protein levels, which resulted in a decrease of the cell proliferation, MMP-2 and MMP-9 activities and invasive potential of Panc-1 cells as well as increased chemosensitivity to Gemcitabine in vitro and tumorigenicity in nude mice.Overall, these results suggest that CD147 plays an important role in proliferation, invasion and chemosensitivity of human pancreatic cancer cell line Panc-1,indicating that CD147 may be a promising therapeutic target for pancreatic cancer.
