目的 改良体外分离培养、扩增、鉴定大鼠毛囊干细胞(rHFSCs)的方法,观察其生物学特性,并在特定条件下诱导分化,探讨其成脂成骨分化能力。方法:无菌条件下切取1周龄SD大鼠触须部皮肤,用Dispase酶和Ⅳ型胶原酶混合液消化,显微镜下分离毛囊隆突部,用梯度加液组织块贴壁法培养rHFSCs,细胞融合达80%时消化传代,用Ⅳ型胶原差速贴壁法纯化。
