mGluR5-PLCbeta4-PKCbeta2/PKCgamma pathways in hippocampal CA1 pyramidal neurons in pilocarpine model

  In pilocarpine induced status epilepticus (PISE) in metabotropic glutamate receptor 5 (mGluR5) knock-out (mGluR5-/-) and wild-type (mGluR5+/+) mice,we showed that at 2 h during PISE,induced PKCbeta2 or PKCgamma immunopositive product was demonstrated at the border between the stratum oriens and alveus (O/A border) of CA1 area in mGluR5+/+ mice.However,much less staining was observed in mGluR5-/-mice.At 1d after PISE,induced PLCbeta4,PKCbeta2 or PKCgamma in stratum pyramidale of CA1 area in mGluR5+/+ but not mGluR54-mice was found.Double labeling showed co-localization of induced PKCbeta2 or PKCgamma with mGluR5 or induced PLCbeta4 in the stratum pyramidale of CA 1 area.It therefore suggests that induced PLCbeta4-PKCbeta2/PKCgamma at 1 d after PISE in pyramidal neurons or induced PKCbeta2 or PKCgamma at O/A border at 2 h during PISE may be involved in mGluR5-related signal transduction leading to delayed loss of pyramidal neurons in mGluR5+/+ mice.
