Objective:To obsreve the effect of Storax on the brain and blood concentration of sulpiride in rats and to explore the possible interaction between sulpirde,a western medicine,and Storax,one of Traditional Chinese medicine,when used together,and to prove the mechanism-whether could Storax raise the permeability of gastrointestinal barrier (GB) and blood-brain barrier and hope that the combination application could reduce the dosages of sulpirde when the drugs administratied.Methods:The rats were randomly assigned into three groups:Storax group (group Ⅰ):Storax(400 mg·Kg-1,p.o.) was administered.sulpiride group (group Ⅱ) sulpiride(400 mg·Kg-1,p.o.) was administered Combination group (group Ⅲ):Storax (400 mg·Kg-1,p.o.)+sulpiride (400 mg·Kg-1,p.o.) was administered.The Microdialysis fluid were collected from corpora striatum area and blood of rats by microdialysis technology at the time periods of(30,60,90,120,150,180) min after oral administration for a week.HPLC (AlltimaC 18 column,250 mm×4.6 mm,5 μm,with a mobile phase consisting of acetonitrile:50 mM KH2PO4)was used to determin concentration of sulpiride in mocrodialysis fluid.The fluorescence detector emission wavelength was set at 238mn and absorption wavelength 345 nm.The detection limit of sulpiride was10ng·mL-1.The in vitro recovery of microdialysate for sulpiride were 42.21%% in artificial cerebrospinal fluid and were 27.18% in Vascular perfusate fluid,respectively.Results:The concentration of sulpiride in rat brain and blood were higher in group Ⅲ than group Ⅱ significantly.The averaged blood concentration of sulpiride in group Ⅲ was 5.577μg · mL-1,and group Ⅱ 1.861 μg · mL-1.The averaged brain concentration of sulpiride in group Ⅱ was 0.2161 μg · mL-1,and in group Ⅱ 0.0955 μ g · mL-1 The average blood-brain ratio was 1∶0.051 in group Ⅱ,and Ⅰ∶ 0.039 in groupⅢ.Conclusion:The concentration of sulpiride in the blood and in the cerebrospinal fluid were increased by combinating utilization of Storax.The fact that Storax can increase the sulpiride concentration of the cerebrospinal fluid (CSF) suggested that Storax can increase the permeation of gastrointestinal barrier and BBB and elevate the concentration of sulpiride in the blood and in the cerebrospinal fluid.
