具有光学活性的环状胺类化合物广泛存在于许多有生物活性的药物和天然产物中.亚胺还原酶是一类近期出现的能催化环状亚胺不对称还原来获取手性胺的生物催化剂.在本研究中,我们发现了一个来自乳酸类芽孢杆菌的亚胺还原酶PISIR.这个酶有很高的活性和热稳定性,在30℃和pH 6.0的条件下测得该酶对底物2,3,3-三甲基假吲哚的纯酶比活高达15.1U/mg,在50℃下测得该酶的半衰期为49.5小时.动力学研究表明PISIR对底物2,3,3-三甲基假吲哚的催化效率常数(kcat/Km)相比于其他报道的亚胺还原酶高很多,达到了23.4 s-1 mM-1.因此,我们首次开发了一条高效还原3H吲哚和3H吲哚亚胺离子的生物还原途径,成功地合成了15种高对映选择性的氮原子上无保护或有保护的S型吲哚啉.
