Given the heterogeneity of the cancer mieroenvironment, it is necessary for therapeutic nanoparticles to consider engineered and rapid throughput approaches for cancer treatment.To illustrate this approach,we are using two cancer models, namely a (i) human multiple drug resistant (MDR) breast cancer and (ii) human pancreatic ductal adenocarcinoma (PDAC) model in nude mice.In the first example, the importance of MDR in the MCF-7 breast cancer line demonstrates targeting of acquired "pump" and "non-pump" resistance mechanisms by a multifunctional mesoporous silica nanoparticle (MSNP) platform.This 50 nm polyethyleneimine-polyethylene glycol (PEI-PEG) copolymer-coated MSNP have been sequentially optimized for systemic drug delivery, siRNA delivery, and dual drug drug/siRNA delivery.
