目的:分别分离培养胰腺干细胞和胰岛微血管内皮细胞,采用聚乳酸-羟基乙酸共聚物(poly(lactic-co-glycolic acid),PLGA)膜片培养细胞。通过胰腺干细胞和胰岛微血管内皮细胞体外共培养,探讨微血管内皮细胞对PLGA的生物改性以及诱导胰腺干细胞向胰岛内分泌细胞分化的作用。方法:选择成年Wistar大鼠麻醉后切取胰腺,用Hanks液洗涤后切碎,P型胶原酶消化,不连续密度梯度离心纯化胰腺干细胞。同时应用免疫磁珠法分离培养胰岛微血管内皮细胞。通过检测内皮细胞标志物第Ⅷ因子相关抗原(vWF)和CD34的表达而确定微血管内皮细胞。
