Statistical experimental designs provided by Design-Expert software were applied to optimize the extraction technology of total tannins from G.carolinianum L..A "Box-Behnken" Design was employed to evaluate the effects of different components in the medium.A series of singlefactor experiments were conducted to investigate the effects of extraction time, methanol concentration, solid-liquid ratio and extraction times on the yield of total tannins.Linear or no-linear mathematic models were used to estimate the relationship between independent and dependent variables.Determined the contents of total tannins in G.carolinianum L.which were collected from different places by the optimum extraction process.The optimal values of the above parameters were the concentration of Methanol of 47%, supersonic extraction time of 45 min, and the solid-liquid ratio of 52: 1.Under the optimized conditions, the experimental value of total tannins was 10.81%.The range of total tannins content of G.carolinianum L.from different areas was 4.42~10.29%.The total tannins content difference to be significantly different regions, The factors of areas should be fully considered when used as medicine.
