Objective: The aims of this study were: (1) to determine whether fragments of a mesenchymal stem cell (MSC) sheet can be used to engineer an injectable bone tissue precursor; (2) to analyze the effects of platelet-rich fibrin (PRF) on MSCs in vitro; and (3) to investigate in vivo bone formation resulting from the injection of MSC sheet fragments with PRF membrane fragments.Methods: We first prepared MSC sheets using a continuous culture method and prepared PRF from the same donors.Then we investigate the effects of PRF on MSCs in vitro by 3-(4,5)-dimethylthiazol (-z-y 1)-3,5-diphenytetrazolium bromide (MTT) assays and on the capacity of MSC sheet in vitro by the expression of alkaline phosphatase (ALP) activity and osteogenesis-related genes (i.e., alkaline phosphatase, osteopontin, osteocalcin, bone morphogenetic protein-2, and collagen type Ⅰ), respectively.And last We injected MSC sheet fragments with or without PRF membranes subcutaneously into severe combined immunodeficiency (SCID) mice to test the effect of PRF on the capacity of MSC sheet in vivo by micro-computed tomography scans(Micro-CT) and histological and histomorphometrical examinations at six weeks after transplantation.
