Aims To determine the toxicity effects of mangiferin on animals.Methods Acute toxicity test in mice and Beagle dogs given maximum-tolerated dose were used to observe the acute toxicity of mangiferin.Long-term toxicity test in rats and Beagle dogs given mangiferin for 30 days were adopted for long-term toxicity.Mutagenicity was observed by the microbial reverse mutation assay,chromosome aberration test,and micronucleus test.Genotoxicity was observed by fertility and early embryonic development toxicity test,rat embryo-fetal developmental toxicity test,rabit embryo-fetal developmental toxicity test.Results 1.The MTD of mangiferin in mice was 54g/kg,and not found significant toxicity in this dose.The MTD of mangiferin in Beagle dogs was 9g/kg,and total bilirubin was significantly higher in one-quarter dogs in this dose,but can self-healing.2.There were no symptoms of toxic reactions in the long-term toxicity study,and there were no toxicological changes in hematology and blood biochemical examination.After given mangiferin with the concentration of 0.32g/ml for 30 days,some rats appeared mechanical intestinal obstruction,further appeared abdominal distension,death and other symptoms,but if reduced the concentration of mangiferin,the phenomenon also decreased.The safety dose of mangiferin in rats was 2.4g/kg,approximately equal to 76 times of the clinical intended dose.The toxicity target organ was digestive canal.The abdominal distension,death and other symptoms were not found in Beagle dogs.3.Genetic toxicity and reproductive toxicity tests were negative.Conclusions Mangiferin is belonging to low toxic compound,without mutagenesis and genetic toxicity,safe in utilization,and have broad prospects for development and application.
