Anti-inflammatory effect of dietary selenium in LPS-induced model of mastitis mouse

  Mastitis is a major cause of economic losses to the dairy industry.Selenium (Se), a biological trace essential element, benefits health and immunity of the mammary gland.In the present study, we investigate that effects of dietary selenium on LPS-induced models of mammary mastitis in mouse.Eighty female BALB/c mice were randomly divided into three groups and fed with daily rations that contain 0.087 mg/(kg· BW), 0.15 mg/(kg·BW) and 1.5 mg/(kg· BW) sodium selenium.At the end of experiment, an intramammary infusion of LPS was used to establish models of mastitis in mouse.Histopathological analysis, enzymatic activity detection and expression of pro-inflammatory genes were conducted to study effects of dietary selenium on LPS-induced models of mammary mastitis in mouse.Results showed that selenium can affect the activity of glutathione peroxidase and mRNA expression of GPX1 and GPX4 of mammary gland in a dose dependent manner.Histopathological observation of mammary gland in models of mammary mastitis in mouse showed more infiltration of inflammatory cells.mRNA expression of TNF-α, IL-1β, IL-6, COX-2 and iNOS of mammary gland of Se-deficient group were higher than Se-adequate group.These results showed that Se can affect LPS-induced inflammatory responses of mammary gland.
