A brief survey of the optimization of the load and wash steps for amentoflavone from podocarpus nagi with macroporous adsorption resin is given in this study.Based on the comparison of several common resins,AB-8 resin with the preferable adsorption and desorption performances was elected in the following tests.The concentration of amentoflavone in loading solution,and eluent,load rate and volume as well wash rate and volume were identified as the critical parameters for the process.And an optimized process was recommended as follows: The concentration,the load rate and volume of loading solution were 0.0324 mg·mL-1,1 BV·h-1 and 9 bed volume (BV),respectively;Desorption was sequentially operated with 80% ethanol-water (v/v) of 3BV of eluent at the load rate of 1 BVh·-1.And the content of amentoflavone has increased from 0.1% to 2.0% with the recovery of 68.58%.The purification process provides a basis for the further development of amentoflavone in podocarpus nagi.
