Backgroud: Neuroimaging studies have shown that major depressive disorder(MDD)is accompanied by functional abnormalities in amygdala and related connections,yet,little is known about the resting-state dynamic functional connectivity(dFC)of amygdala subregions(including basolateral,BL; centromedial CM; superficial,SF)alterations with the whole-brain networks in first-episode,untreated MDD patients.Methods: In a cross-sectional study,30 first-episode,untreated MDD patients and 63 healthy controls(HC)underwent gradientecho echo-planar imaging scan on a 3-T MR system.Six subregions of bilateral amygdala were selected as seed areas.Onesample t tests and general linear model analysis were used to assess the within-group dFC patterns and between-group differences of the mean strength and variance of dFC of each amygdala subregion with the gray matter by using age and gender as covariates.Results: The within-group analysis revealed similar spatial dFC pattern of bilateral amygdalar subregions in the MDD and HC groups.Compared with controls,MDD patients showed decreased positive mean dFC of bilateral CM and SF with brainstem and cerebellum as well as left SF and left thalamus(corrected P<0.05).MDD patients showed decreased negative mean dFC of left CM with right superior frontal gyrus and right CM with right medial temporal gyrus(corrected P<0.05)when compared with controls.Additionally,the MDD-HC group differences in mean positive dFC strength between bilateral CM and cerebellum showed positive correlation with onset age and MDD-HC group difference in mean dFC between right SF and cerebellum as well as MDDHC group difference in mean positive dFC variability between right CM and left SMA were positively associated with disease duration of MDD.Conclusions: Our findings confirmed the distinct function of amygdala subregion and the limbic-thalamo-cortical circuitry played a crucial role in MDD.
