  Apoptosis induced by microbial infections are factors implicated in the pathogenesis of Helicobacter pylori (H.pylon) infection.Here we show that human gastric epithelial cells sensitized to H.pylori confer susceptibility to TRAIL-mediated apoptosis.Human gastric epithelial cells were resistant to TRAIL despite expression of TRAIL death receptors and induction of caspase-8 activation after TRAIL engagement.However,in the presence of H.pylori,TRAIL induces sequential activation of caspase-8,Bid cleavage,activation of mitochondrial pathway,leading to breaking apoptosis resistance.The H.pylori-induced TRAIL sensitivity could be blocked by capsase-9 inhibitor and overexpression of Bcl-2,indicating mitochondrial pathway is required for apoptosis signaling.Therefore,the induction of TRAIL sensitivity by H.pylori is dependent on activation of caspase-8 downstream pathway to convey the death signal to mitochondria,leading to activation of mitochondrial signaling pathway and breaking apoptosis resistance.Modulation of host apoptosis signal transduction by bacterial interaction adds a new dimension to the immune pathogenesis in Helicobacter infection.
