microRNAs (miRNAs)是一类长度为18~23 nt的非编码小RNA,在转录后水平调控基因的表达,功能上涉及细胞分化、增殖、凋亡、胚胎发育和器官形成等过程.2012年,我们报道了一种灵敏特异的miRNA检测技术——S-Poly(T)法(Kang,PLoS One, 2012),本工作描述其升级版S-Poly(T) plus法.S-Poly(T) plus法将miRNA的多聚腺苷酸化及反转录两个步骤合为一步,不仅简化步骤,还使灵敏性提高4~8倍,并具有极优良的线性关系.通过引入一种核酸助沉剂miRCOP,miRNA回收效率明显优于市面上常见提取试剂盒,特别适合从低丰度样本(如血清)中提取miRNA.
