AIM To investigate protect effect of Fufangdengzhanhua dripping pill (FDD)s oxidative stress induced by hydrogen peroxide on PC12 cell.METHODS PC12 cells were divided into six groups in culture: control group, oxidative damage group,different concentration of FDD group (0.763, 3.052, 12.207 mg· L-1), positive drug group (vitamin E, 4.307 mg· L-1).After drug intervention for 24 h, PC12 cells were injured by 300 μmol·L-1 H2O2 except control group for 2 h.With the pretreating of FDD at different concentration for 24 h, typical morphological characteristics of PC12 after being exposed to H2O2 could be observed under the inverted microscope.The survival rate of PC12 were detected by MTI.Lactate dehydrogenase (LDH), superoxide dismutase (SOD)and malondialdelyde (MDA) in the culture medium.RESULTS After being exposed to 300 μμnol· L-1 H2O2 for 2 h, FDD can decrease LDH, MDA content and increase SOD activity significantly.FDD increases the survival rate of PC12 cells according to the results of MTT.Compared with the model group, there was statistically significant difference (P <0.01).CONCLUSION FDD can protect injury induced by hydrogen peroxide on PC12 cells in culture.The acting mechanisms may include protecting cell membrance and inhibiting lipid peroxidation.
