BACKGROUD: The treatment of chronic wound resulted from diabetes has been reported by a lot of researchers used stem cells to promote wound healing.However, scar formation and non-appendages healed were still in previous studies.METHODS: We divided mice into four groups: diabetes treatment group, diabetes control group, normal treatment group and normal control group.After creation of wound, we transplanted embryonic skin cells which were from E17.5-18.5 mice embryos into wound area.Gross observations were made after 4, 7, 11 and 14 days.And histological observations were made after 1, 2 and 3 weeks.Epidermal and dermal regeneration degree, granulation tissue thickness, angiogenesis, collagen orientation and quantity of hair follicles were calculated.Transforming growth factors beta (TGF-β) were detected by immunohistochemistry.RESULTS: The wound closure rate of diabetes control group was significant slower than the normal control group at each check point (p<0.01).The histological scores of diabetes treatment group were similar with normal treatment group but better than diabetes control group significantly (p<0.05).Collagen orientation evaluated by fast Fourier transformation was similar between diabetes treatment group and normal treatment group.We counted hair quantify of diabetes treatment group and normal treatment group, no significant differences between them, p=0.36.TGF-β1 and 3 were expressed in regenerative skin in different phase.CONCLUSION: Our study indicated that embryonic skin cells have ability to repair the skin defect of diabetes with regenerative skin and appendages whilst no scars formation.It perhaps provides a better way to cure the chronic wounds without scars.
