Vmhxkl, a pathogenicity-related gene predicted to encode hexokinase, was obtained by amplifying the flanking sequence of T-DNA of one pathogenicity weakened mutant.Vmgxk1, encoding glucokinase, another gene of hexokinase gene family was acquired by the genomic information of V.mali.In this study, the two gene Vmhxk1 and Vmgxk1 function were analyzed by gene knockout and complementation assays.The result showed that Vmgxk1-deficient mutants were indistinguishable from the wild-type in all growth parameters tested except the time of conidia formation which was delayed for 15 days.In contrast, △Vmhxk1 mutants lacking Hxk1 showed a pleiotropic growth defect.On artificial media, vegetative growth was retarded, and the time of conidia formation postponed very late.Compared with the wild type 03-8, the pathogenicity of △Vmhxk1 mutants was weakened.The complementation of △Vmhxk1 and △Vmgxk1 mutants showed similar phenotype and pathogenicity to 03-8.The results indicated that Vmgxk1 was involved in the conidia formation and Vmhxk1was not only related to conidia formation, but also mycelium growth and pathogenicity.
