基底力学微环境对干细胞生物学行为的影响

  干细胞是一类具有自我复制和分化潜能的细胞,可分化成多种功能细胞,其生物学行为(如表型、生长、迁移、增殖和分化)在很大程度上受到细胞内源和外源因素的共同调节,而力学微环境和细胞的粘附接触对于干细胞的自我更新和命运的抉择是非常重要的。为模拟体内二维壁龛微环境、考察不同基底硬度和/或拓扑结构对干细胞生物学特性及干性和分化的影响,本文制备了不同硬度、不同拓扑结构及二者耦合的聚丙烯酰胺水凝胶(PA胶)基底,并建立大鼠骨髓间充质干细胞(rMSCs)及无饲养层细胞培养条件下小鼠胚胎干细胞(mESCs)在PA胶上的正常生长体系。
