Vibrio parahaemolyticus is a major pathogen frequently involved in human outbreaks of seafood-associated gastroenteritis.The aim of this study was to use two PCR-based techniques for the analysis of genetic variability among V.parahaemolyticus virulence strains isolated from seafood in Shanghai from 2012 to 2013.All isolates were analyzed for the presence of tdh and trh gene by PCR.48 strains(tdh+) of V.parahaemolyticus isolated from shrimps were analyzed by repetitive extragenic palindromic PCR (REP-PCR) and renterobacterial repetitive intergenic consensus sequence PCR (ERIC-PCR).Results indicated that 48 strains of V.parahaemolyticus,representing a wide range of patterns,were grouped into 5 and 3 clusters at a similarity coefficient of 0.66 by REP-PCR,and ERIC-PCR,respectively.ERIC-PCR is preferable to REP-PCR because of the greater discriminative abilities of its fingerprints,while REP-PCR may be a practical method because it generates fewer amplification bands and patterns.Thus,this study demonstrated the genetic variability of V.parahaemolyticus (tdh+) in shrimp,provided insights into the dissemination of virulence strains,and improved our knowledge of methods of microbiological risk assessment in seafood from market.
