Aim To investigate the different pharmacokinetic behavior of 58Fe after oral administration ofhemin in normal rats and the iron deficiency rats.Methods A rapid, sensitive, and specific ICP-MS method for the determination of 58Fe in rat plasma was developed and a rat model of iron deficiency anemia was established.Noncompartmental method was used in the calculation of pharmacokinetic parameters.Results It was found that hemin absorption increased significantly under iron deficiency anemia status,with the AUC0-t, AUC0-∞, and Cmax showed a over 2-fold rise.Besides, the CLz and MRT0-t of iron deficiency rats also showed a significant difference from normal rats.Conclusion Iron deficiency status can significantly increase hemin absorption, and this study indicated that receptors and transporters involving in iron uptake, such as divalent metal transporter 1 (DMT 1) and herne carrier protein 1 (HCP1), may up-regulated under the status of iron deficiency anemia.
