Erythropoietin (EPO) is an important therapeutic protein and can be also used as doping reagent in sports.Due to the presence of a large number of glycoprotein isoforms, the slight difference in their physicochemical properties, the trace concentration in urine and blood, and the complexity of sample matrix, a specific enrichment, high resolution separation and sensitive detection method is important in EPO analysis.Herein, we present a hyphenated approach of immuno-magnetic beads-based extraction (IMBE) and capillary zone electrophoresis (CZE) with deep UV laser-induced fluorescence (deep UV-LIF) detection for the enrichment, separation and detection of EPO.The IMBE step permitted convenient extraction of EPO from sample matrix.The CZE separation allowed for high-resolution profiling of the glycoforms of EPO within 25 min.
