Polygonum cuspidatum [1],a traditional Chinese herb,is rich in resveratrol,but mostly exists in glucosides with glucosidic bond.Enzyme preparations can hydrolyze and degrade the cell wall rendering the intracellular materials more accessible for extraction and can break down glucosidic bond and enzyme-assisted extraction is a method which possesses the advantages of environmental-fiiendship,high efficiency and easy operation process [2].Then a systematic study need to be carried out on emzyme type.Classical optimization studies used the one-factor-at-a-time approach which is time-consuming and expensive.Uniform design (UD) which just needs far fewer trials is widely accepted [3].So Enzyme-assisted extraction of resveratrol from polygonum cuspidatum was comprehensively investigated by uniform design optimization in this work.The effect of varying different parameters such as enzyme concentration,extraction time,pH of enzyme solution and extraction temperature on the yield of resveratrol was optimized.The results showed the best experimental conditions were as follows:the concentration of the enzyme was 29.7% (w/w),extraction temperature was 45 ℃,pH value of enzyme solution was 3.0 and extraction time was 8.13 h.Under the optimum conditions,the extraction yield of resveratrol showed a remarkable increase (624%),compared with the untreated one.
