本文研究草棉(Gossypium Herbaceum)花花瓣两种提取工艺产物(聚Ⅰ和聚Ⅱ)对小鼠凝血时间、大鼠血小板粘附功能、血小板聚集功能及大鼠动-静脉旁路血栓形成的影响。采用玻片法体外测定小鼠全血凝血时间,以阿司匹林(Aspirin,24. 7 mg/kg,ig)和丹参滴丸(Salvia Miltiorrhiza,105. 3mg/kg,ig)为阳性对照,聚.Ⅰ和聚Ⅱ分别分为35 mg/kg、70 mg/kg两个剂量组。结果聚Ⅰ(35mg/kg,ig)能明显缩短凝血时间(P<0. 05) 。其余剂量组对凝血时间均无明显影响。采用胶原粘附法测定大鼠血小板粘附功能,以血小板粘附率为观察指标,以阿司匹林(17. 1mg/kg,ig)和丹参滴丸(72. 9 mg/kg,ig)为阳性对照,聚Ⅰ和聚Ⅱ分别分为25 mg/kg、50 mg/kg、100 mg/kg三个剂量组,结果聚Ⅰ(25mg/kg、50 mg/kg,ig)、聚Ⅱ(25mg/kg,ig)均能增强大鼠血小板粘附功能(P<0. 05) 。采用比浊法测定大鼠血小板聚集功能,分别以二磷酸腺苷(Adenosine-5’-Diphosphate,ADP,终浓度为3. 33umol/1) 和胶原(Collagen,终浓度为212ug/1) 诱导血小板聚集,阿司匹林(17. 1mg/kg,ig)和丹参滴丸(72. 9 mg/kg,ig)为阳性对照,以最大聚集率为观察指标,结果聚Ⅰ(50 mg/kg、100 mg/kg.ig)、聚Ⅱ(50 mg/kg、100 mg/kg,ig)对ADP诱导的大鼠血小板聚集有明显抑制作用(P分别<0. 05和0. 001) ,抑制率分别为16. 79%、21. 98%和11. 71%、17. 67%,呈剂量依赖性。聚Ⅰ(100 mg/kg,ig)对Col诱导的血小板聚集有明显抑制作用(P<0. 05) 。抑制率为13. 35%。聚Ⅱ(25mg/kg,ig)使Col诱导的血小板聚集的潜伏期明显缩短,潜伏期为1分钟,生理盐水组的潜伏期为3分钟,而阿司匹林组的潜伏期为5分钟。在大鼠动-静脉旁路血栓模型中,聚Ⅰ(50 mg/kg,ig)和聚Ⅱ(50mg/kg,ig)均能使血栓湿重明显减轻(P<0. 05) 。血栓形成抑制新抢医科大学医学硕士学位论文率分别为4229%和29.17%。结论:聚I较聚n抗栓作用强,可在确定聚I提取工艺的基础上进一步优化。关键词草棉;凝血时间;血小板粘附;血小板聚集; 动一静脉旁路血栓The effeet 5 of GossyPium Herbaeeum extraet on Platelet aggregation and arteriovenous shunt model in rats ABSTRACT:In this article,we studied the effeets of GossyPiumHerbaeeum extraet,Extraet A and ExtraetB,PrePared in two extraetionmethods,on coagulation time in miee and Platelet aggregation andPlatelet adhereranee and the formation of thrombosis in arteriovenousshunt model in rats,resPeetively.Extraet A and Extract B were given atthree doses(25 mg/kg、50 mg/kg、100 mg/kg)and AsPirin(24.7 mg/kg,19)and Salvia Miltiorrhiza(105.3mg/kg,19)were used as Positive eontrols toevaluate whole eoagulation time by glass slide method and the resultsshowed that Extraet A at a dose of 25mg/kg,1.9.markedly deerease theeoagulation time(P<0.05)while other doses having no significanteffeets on eoagulation time.Platelet adheranee funetion was assessed byusing eollogen adheranee method with Platelet adheranee rate as the index.EXtract A and Extraet B Were given at three doses(25 mg/kg、50 mg/kg、100mg/kg)with AsPirin(17.lmg/kg,19)and Salvia Miltiorrhiza(72.9mg/kg,19)as the Positive eontrols.The results showed that both ExtraetAat a dose of 25mg/kg and 50 mg/kg,19 and Extraet B at at a dose of25mg/kg,19 augmented the Platelet adheranee funetion in rats(P<0.05);Turbidity method was used to assess the Platelet aggregation funetion in一2一
