多叶姜黄为姜科姜黄属植物Curcuma comosa Roxb.的根茎,生于热带、亚热带,广泛分布于泰国,印度尼西亚及马来西亚。为泰国传统用药,在当地被广泛用于治疗产后子宫出血而引起的炎症。本课题组前期的研究结果表明,该植物的甲醇提取物具有较强的抗炎活性,为明确其抗炎作用的物质基础,对多叶姜黄甲醇提取物的化学成分进行了系统研究,利用硅胶开放柱色谱,ODS开放柱色谱,及HPLC等分离手段从中分离得到了60个化合物,通过IR,UV,NMR,MS,CD等光谱手段,结合化学衍生法,确定了56个化合物的结构,其中倍半萜类化合物31个,分别为:comosoneⅠ(1),comosoneⅡ(2),dimethoxycurcumenone(3),comosoneⅢ(4),莪术烯酮/姜黄烯酮(curcumenone,5),comosol(6),curcolonol(7),蓬莪术烯酮(curzerenone,8),异蓬莪术环二烯酮(isofuranodienone,9),蓬莪术环二烯酮(furanodienone,10),1(10)Z,4Z-蓬莪术二烯-6-酮(1(10)Z,4Z-furanodiene-6-one,11),吉马酮/牻牛儿酮(germacrone,12),13-羟基吉马酮(13-hydroxygermacrone,13),吉马酮二环氧化物(germacrone-1(10),4-diepoxide,14),去氢姜黄二酮(dehydrocurdione,15),新姜黄二酮(neocurdione,16),姜黄二酮/莪术双酮(curdione,17),7α-羟基新姜黄二酮(7α-hydroxyneocurdione,18),7β-羟基姜黄二酮(7β-hydroxycurdione,19),蓬莪术环氧酮(zederone,20),蓬莪术环氧酮环氧化物(zederone epoxide,21),欧亚活血丹内酯(glechomanolide,22),泽泻醇(alismol,23),异原莪术烯醇(isoprocurcumenol,24),原姜黄环氧奥烯醇(procurcumenol,25),aerugidiol(26),zedoalactone B(27),zedoarondiol(28),isozedoarondiol(29),环氧泽泻烯(alismoxide,30),curcumadione(31);酚类化合物6个,分别为:4-(3,5-二羟基己基)苯酚[4-(3,5-dihydroxyhexyl)phenol,32],对羟基苯甲醛(4-hydroxybenzaldehyde,33),香草醛(4-hydroxy-3-methoxybenzaldehyde,34),覆盆子酮[4-(4-hydroxyphenyl)butan-2-one,35],杜鹃醇[(+)-rhododendrol,36],4,6-二羟基-2-O-(β-D-吡喃葡萄糖基)苯乙酮[4,6-dihydroxy-2-O-(β-D-glucopyranosyl) acetophenone,37];单萜及单萜苷类7个,分别为:comosoxide A(38),comosoxide B(39),comososide(40),2,2,6-trimethyl-1-oxaspiro[2,5]oct-5-en-4-ol(41),1-hydroxy-α,α,4-trimethyl-3-cyclohexene-1-methanol(42),6-hydroxy-3(1-hydroxy-1-methylethyl)-6-methyl-2-cyclohexen-1-one(43),(1S,2S,4R)-1,8桉叶素-2-O-β-D-葡萄糖苷[(1S,2S,4R)-2-hydroxy-1,8-cineoleβ-D-Glucopyranoside,44)];二芳基庚烷类化合物12个,分别为(3S',5S')-3,5-二乙酰氧基-1-(3,4-二羟基苯基)-7-(4-羟基苯基)庚烷[(3S',5S')-3,5-diacetoxy-1-(3,4-dihydroxyphenyl)-7-(4-hydroxyphenyl)heptane,45],(3S',5S')-3,5-二羟基-1-(3,4-二羟基苯基)-7-(4-羟基苯基)庚烷[(3S',5S')-3,5-dihydroxy-1-(3,4-dihydroxyphenyl)-7-(4-hydroxyphenyl)heptane,46],(3R',5S')-3,5-二羟基-1-(3,4-二羟基苯基)-7-(4-羟基苯基)庚烷[(3R',5S')-3,5-dihydroxy-1-(3,4-dihydroxyphenyl)-7-(4-hydroxyphenyl)heptane,47],(3S',5S')-3-乙酰氧基,5-羟基-1,7-双(4-羟基苯基)庚烷[(3S',5S')-3-acetoxy,5-hydroxy-1,7-bis(4-dihydroxyphenyl) heptane,48],(3S',5S')-3,5-二乙酰氧基1,7-双-(4-羟基苯基)庚烷[(3S',5S')-3,5-diacetoxy-1,7-bis(4-hydroxyphenyl)heptane,49],(3R,5S)-m-3,5-二羟基-1,7-双(3,4-二羟基苯基)庚烷[(3R',5S')-3,5-dihydroxy-1,7-bis(3,4-dihydroxyphenyl)heptane,50],(3R',5S')-3,5-二羟基-1-(3-羟基-4-甲氧基苯基)-7-(3,4-二羟基苯基)庚烷[(3R',5S')-3,5-dihydroxy-1-(3-hydroxyl-4-methoxyphenyl)-7-(3,4-(dihydroxyphenyl) heptane,51],(3S',5S')-3,5-二乙酰氧基-1,7-双-(3,4-二羟基苯基)庚烷[(3S',5S')-3,5-diaceoxy-1,7-bis(3,4-dihydroxyphenyl)heptane,52],(3S',5S')-3,5-二羟基-1,7-双(3,4-二羟基苯基)庚烷[(3S',5S')-3,5-dihydroxy-1,7-bis(3,4-dihydroxyphenyl)heptane,53],(3R',5S')-3,5-二羟基-1-(3,5-二甲氧基-4-羟基苯基)-7-(4-羟基苯基)庚烷[(3R',5S')-(3,5-dihydroxy-1-(3,5-dimethoxy-4-hydroxyphenyl)-7-(4-hydroxyphenyl)heptane,54],(5S)-5-羟基-1,7-双(4-羟基苯基)-3-庚酮[(5S)-5-hydroxy-1,7-bis(4-hydroxyphenyl)-3-heptanone,55],1,7-双(4-羟基苯基)-4E,6E-庚二烯-3-酮[1,7-bis(4-hydroxyphenyl)hepta 4E,6E-dien-3-one,56]。其中新化合物16个,分别为化合物1-4,6,32,38-40,45,47-51,54;新天然产物1个,为化合物41;属中首次分离的化合物5个,分别为化合物11,23,42-44;已知化合物除37外均为种内首次分离得到。
