建立高效、稳定的再生体系是果树转基因成功的关键环节,为此,本研究以中国沙棘,俄罗斯大果沙棘为试验材料,就影响其植株再生的有关因子,包括植物生长调节剂种类及浓度、暗培养时间、叶片极性、基本培养基、外植体类型、糖源种类及浓度等多方面进行研究探讨,建立了沙棘高效离体再生体系。主要结果如下:1中国沙棘诱导体胚发生以1/2B5为基本培养基,附加30g/L蔗糖再生效果最好。田间叶片再生体胚的最适培养基为1/2B5+6-BA3.0mg/L+NAA0.1mg/L;试管苗叶片、体胚苗叶片再生体胚的最适培养基为1/2B5+6-BA2.0mg/L+NAA0.1mg/L;子叶再生体胚最适培养基为1/2B5+6-BA0.5mg/L+NAA0.05mg/L。2暗试验结果表明,子叶接种后进行一段时间的暗培养(2-3周),可明显提高沙棘子叶体胚再生率。3子叶极性试验结果表明,子叶近远轴端均可产生体胚,但近轴端再生率高于远轴端;体胚绝大多数在子叶近轴面产生。4子叶、叶片、胚轴、茎段在其适宜培养条件下,子叶再生率最高。5培养基中附加LH对离体子叶再生有抑制作用。6试管苗生根培养基以B5+6-BA0.05mg/L+NAA0.5mg/L效果最好。
