Flavonoids and phenolic acids were two important natural products in plants,which had antioxidant,anti-neoplastic and anti-cardiovascular and cerebrovascular diseases and other functions.the two natural products showed promising prospects,but the plant source was limited and the using level was low,consequently developing new plant sources and other purposes was of great significance.This work was carried out to study the extraction,purification and microbial transformation process of flavonoids and phenolic acids in Rubus Suavissmus S.Lee and phenolic acids in Fatsia japonica(Thunb.)Decne.et Planch..The main results were as follows:The influence of different factors on the extraction yield of flavonoids in Rubus Suavissmus was investigated.The extracting condition was as follows:the ethanol concentration was 70%,the liquid-solid ratio was 40:1,the extraction time was 50 min and extracted 2 times,the extraction yield was 91%.The negtive rlectrospary ionization tandem mass spectrometry method was used to identifify the ingredients of Rubus Suavissmus,two phenolic acids were identified as 3,5-dihydroxy-4-methoxybenzoic acid and gallic acid.Six kinds of flavonoids were identified as:1-feruloyl-P-D-glucose,vitexin,rutin,hyperoside,quercitrin,myricetin-3-O-β-D-gluconside,quercetin(3,5-dihydroxy-4-methoxybenzoic acid,vitexin,myricetin-3-O-β-D-gluconside were reported for the first time).The results showed that in the separation and purification process of Rubus Suavissmus,macroporous resin AB-8 had the best adsorptive effect among eight kinds of macroporous adsorption resins.500 g Rubus Suavissmus can obtain flavonoids samples 78.2 g(the purity was 34.7%,recovery rate was 59%),phenolic acids samples 6.37 g(the purity was 57.4%,recovery rate was 71%).Further isolation and purification of Rubus Suavissmus by using preparative HPLC showed that:500 g Rubus Suavissmus can get 3,5-dihydroxy-4-methoxybenzoic acid 165 mg(the purity was 88.5%),gallic acid 443 mg(the purity was 93.4%),hyperoside 317 mg(the purity was 91.8%),quercetin 357 mg(the purity was 87.5%),rutin 243 mg(the purity was 85.7%).The study of phenolic acids in Fatsia japonica indicated that:seven kinds of phenolic acids were identified as chlorogenic acid,quinic acid,caffeic acid,5-0-feruloylquinic acid,3,4-dicaffeoylquinic acid,3,5-dicaffeoylquinic acid and 4,5-dicaffeoylquinic acid.The three dicaffeoylquinic acids in Fatsia japonica were found for the first time.The content of phenolic acids in different parts of Fatsia japonica was determined:the content of phenolic acids in tender leaves was the highest among different parts in Fatsia japonica(9.2%),the content of 3,5-dicaffeoylquinic acid accounting for 61.4%of total phenolic acids in tender leaves.The polyamide resin was found to be the most appropriate for the separation of phenolic acids in Fatsia japonica.200 g tender leaves of Fatsia japonica can get dicaffeoylquinic acids 8.45 g(the purity was 34.8%,recovery rate was 57%).The ethyl acetate was used on the extraction and purification of phenolic acids,200 g tender leaves of Fatsia japonica obtained ethyl acetate extracts 28.6 g,which contained 3,5-dicaffeoylquinic acid 3488 mg,4,5-dicaffeoylquinic acid 582 mg.22.6 g extraction raffinate was obtained after freeze-drying treatment,which contained quinic acid 3258 mg.The microbial transformation of Rubus Suavissmus extract showed that:the flavonoids in Rubus Suavissmus can be transformed by Aspergillus niger,Aspergillus aculeatus and Eurotium cristatum.The transformation product was identified as 1-feruloyl-β-D-glucose.The transformation product of Fatsia japonica extract was caffeic acid,which was transformed by Aspergillus niger and Aspergillus aculeatus.The transformation efficiency was the highest by using Aspergillus niger(under the condition of pH 5.0,45℃,the concentration of caffeic acid increased from 0.02 to 0.3 mg/mL,increased by 14 times).
