目的:观察并比较几种常用的看家基因,包括β-actin,GAPDH,18s RNA,cyclophilin A 等在中枢和外周神经损伤后胞体分布区神经组织中表达的变化。方法:Wistar 雄性大鼠随机分为右侧坐骨神经夹伤,右侧坐骨神经结扎和胸段脊髓半横断三组。手术后7d,用相对定量 RT-PCR 的方法检测几种看家基因在受损神经元胞体分布区神经组织中的表达水平。结果:GAPDH 和 Cyclophilin A 的表达在不同模型中的变化幅度较小,损伤与对照侧比较差异均无显著性。β-actin 和18sRNA 的表达在不同模型中变化不同,在脊髓损伤和坐骨神经结扎后表达的变化有统计学意义。结论:本研究结果为研究神经系统损伤后相关基因表达变化时内参照基因的选择提供参考,同时也为进一步认识看家基因更加广泛的功能提供依据。
OBJECTIVE: To observe and compare the expression changes of several commonly used housekeeping genes including β-actin, GAPDH, 18s RNA and cyclophilin A in the distribution of nerve cells in the cell body after central and peripheral nerve injury. Methods: Wistar male rats were randomly divided into three groups: right sciatic nerve injury, right sciatic nerve ligation and thoracic spinal cord hemisection. Seven days after operation, the relative quantitative RT-PCR method was used to detect the expression levels of several housekeeping genes in the neural tissues of the damaged neuronal cell body. Results: The changes of GAPDH and Cyclophilin A in different models were small and the differences between the two groups were not significant. The expression of β-actin and 18sRNA varied in different models, and there were significant differences in the expression of β-actin and 18sRNA after spinal cord injury and sciatic nerve ligation. CONCLUSION: The results of this study provide a reference for the study of the selection of internal reference genes when the expression of related genes changes after nervous system injury, and provide a basis for further understanding of the broader functions of housekeeping genes.
