目的应用实时荧光定量PCR法对灌胃不同配伍比例的附子和红参的大鼠粪便中的总菌、乳杆菌和拟杆菌的相对含量进行检测分析。方法将15只SD大鼠随机分成A、B、C三组,每组5只。A组:附子单独给药7d,B组:附子、红参配伍比例1∶1给药7d,C组:附子、红参配伍比例1∶2给药7d。各组灌胃7d结束之后,于第8天收集大鼠的粪便,提取粪便中细菌的DNA,并根据细菌的16SrDNA基因序列设计总菌、乳杆菌和拟杆菌的种属特异性引物,进行实时荧光定量PCR反应,分析不同细菌的相对含量。结果 C组大鼠中总菌、乳杆菌和拟杆菌的相对含量要显著高于A组和B组;A组和B组中总菌、乳杆菌和拟杆菌的相对含量没有明显差异。结论当附子和红参的配伍比例为1∶2时,能在一定程度上改善肠道的微生态环境,促进有益菌的生长。
Objective To detect and analyze the relative contents of total bacteria, Lactobacillus and Bacteroidetes in the feces of rats with different compatibility of aconite and red ginseng by real-time fluorescence quantitative PCR. Methods Fifteen SD rats were randomly divided into three groups (A, B and C). Group A: aconite alone administered 7d, group B: aconite, red ginseng compatibility ratio of 1: 1 dosing 7d, group C: aconite, red ginseng compatibility ratio of 1: 2 dosing 7d. After the 7th day of gavage in each group, the stools of rats were collected on the 8th day, and the DNA of bacteria in the stool was extracted. According to the 16SrDNA gene sequences of bacteria, the species-specific primers of total bacteria, Lactobacillus and Bacteroidetes were designed, Fluorescent quantitative PCR reaction, analysis of the relative content of different bacteria. Results The relative contents of total bacteria, Lactobacillus and Bacteroidetes in C group were significantly higher than those in A and B groups. There was no significant difference in the relative contents of total bacteria, Lactobacillus and Bacteroides in group A and B. Conclusion When the compatibility ratio of aconite and red ginseng is 1: 2, the intestinal microflora can be improved to some extent and the growth of beneficial bacteria can be promoted.
