剑麻是中国热区最重要的麻类经济作物,中国剑麻主栽品种H.11648经过近五十年的种植,出现了明显的早衰早花现象,给剑麻产业带来了严重的经济损失。为此,我们实验室从坦桑尼亚引进了几株晚花品种(A.sisalana Perr ex.Engelm)。为了将来能开展品种评比实验和杂交育种,需要对该晚花品种进行组培快繁。为此,我们以晚花剑麻吸芽苗的茎尖为外植体,比较了不同消毒方法、植物激素浓度配比对晚花剑麻组培快繁的影响,结果表明:最佳消毒方法为75%乙醇1 min+0.1%升汞30 min;芽增殖最佳培养基为MS+6-BA1.5 mg/L+NAA 0.3 mg/L;生根最佳培养基为MS+IBA 1.5 mg/L。
Sisal is the most important cashmere economic crop in China’s hot zone. After the cultivation of Chinese sisal H.11648 for nearly 50 years, the phenomenon of early premature decay occurred early and brought a serious economy to the sisal industry loss. To this end, our laboratory introduced several varieties of late-flowering (A.sisalana Perr ex. Engelm) from Tanzania. In order to be able to carry out varieties of experiments and hybrid breeding in the future, the need for the late flowering varieties of tissue culture fast propagation. For this reason, we use the tip of late sisal aspiration bud shoot explants, compare the different disinfection methods, plant hormone concentration ratio of Siberian hibiscus tissue culture fast propagation, the results show that: the best method of disinfection The optimum medium for bud proliferation was MS + 6-BA 1.5 mg / L + NAA 0.3 mg / L, and the best medium for rooting was MS + IBA 1.5 mg / L.
