目的:研究姬松茸多糖(ABP-AW1)对OVA(ovalbumin,OVA)抗原诱导的细胞因子特异性转录因子的表达,探讨影响免疫应答的相关信号通路。方法:ABP-AW1和OVA皮下2点免疫ICR小鼠,于1 d,15 d各免疫1次,d 28分离培养脾细胞,免疫鼠脾淋巴细胞体外给予抗原OVA再刺激24 h。RT-PCR法分析ABP-AW1对脾细胞因子IFN-γ,IL-4mRNA表达水平及核转录因子T-bet和GATA-3mRNA的相对表达量,ELISA法检测培养上清细胞因子IFN-γ和IL-4的分泌。结果:ABP-AW1能增强OVA诱导的小鼠脾脏Tbet/GATA-3转录因子和Th特征性细胞因子基因表达以及脾细胞分泌水平的升高。结论:ABP-AW1通过IFN-γ/T-bet信号通路,调节OVA抗原的免疫应答机制。
OBJECTIVE: To study the expression of cytokine-specific transcription factors induced by OVA antigen by ABP-AW1 and to explore the related signaling pathways that influence immune response. Methods: The ICR mice were immunized subcutaneously with ABP-AW1 and OVA twice a day for 15 days. The spleen cells were isolated and cultured at day 28, and splenic lymphocytes were stimulated with antigen OVA in vitro for 24 hours. The relative expression levels of IFN-γ and IL-4 mRNA of splenic cytokines and the expressions of T-bet and GATA-3 mRNA of splenic lymphocytes were analyzed by RT-PCR. The levels of cytokines IFN-γ and IL-4 -4 secretion. Results: ABP-AW1 enhanced OVA-induced spleen Tbet / GATA-3 transcription factor and Th-specific cytokine gene expression and increased secretion of splenocytes. Conclusion: ABP-AW1 regulates the immune response mechanism of OVA antigen through IFN-γ / T-bet signal pathway.
