目的:研究促凋亡基因bak在星形细胞瘤(Astrocy-toma)中的表达与意义.方法:采用兔抗人Bak多克隆抗体及免疫组织化学SABC法.结果:在66例星形细胞瘤中,Bak表达39例(59.1%);10例用于对照的正常脑组织中,Bak阳性仅1例(10%).bak在星形细胞瘤中的表达强于正常对照组(P<0.05),bak在Ⅰ,Ⅱ,Ⅲ,Ⅳ级星形细胞瘤相互之间表达无显著差异(P>0.05),但在31例低度恶性星形细胞瘤(WHOⅠ级~Ⅱ级)中表达强于在35例高度恶性星形细胞瘤(WHOⅢ级~Ⅳ级)中的表达(P<0.05).结论:促凋亡基因bak在大部分星形细胞瘤中有不同程度表达,bak可能参与星形细胞瘤的凋亡调变;其表达与星形细胞瘤分化有关.
4. Objective : To study the expression and significance of proapoptotic gene bak in astrocytoma. Methods: Rabbit anti-human Bak polyclonal antibody and immunohistochemistry SABC method were used. RESULTS: In 66 astrocytomas, Bak was expressed in 39 cases (59.1%); in 10 normal brain tissues used for control, only 1 case was Bak positive (10%). The expression of bak in astrocytoma was stronger than that in normal control group (P<0.05), and there was no significant difference in the expression of bak between astrocytoma I, II, III, and IV (P>0.05). However, in 31 cases of low-grade malignant astrocytoma (WHOI grade -II grade), the expression was higher than that in 35 cases of high-grade malignant astrocytoma (WHO III grade -IV grade) (P<0.05). CONCLUSION: The pro-apoptotic gene bak is expressed in different degrees in most astrocytomas, and bak may be involved in the apoptosis modulation of astrocytoma; its expression is related to the differentiation of astrocytoma.
