目的 探讨高血糖对细胞凋亡的影响以及细胞凋亡与糖尿病肾病发生机制的关系。方法 应用静脉输注葡萄糖造成大鼠体内的高血糖 ,运用琼脂糖电泳及流式细胞术检测大鼠肾脏皮质细胞凋亡及其相关基因蛋白的表达。结果 短时间大量静脉输注葡萄糖后 ,高血糖大鼠肾皮质细胞凋亡增加 ,bc1 2基因表达下调 ,bax、Fas基因表达上调 ,但与输注甘露醇对照组大鼠无显著差异。结论 高渗透压可能为高血糖诱导细胞凋亡的机制之一 ,细胞凋亡与早期糖尿病肾病发生密切相关
Objective To investigate the effect of hyperglycemia on apoptosis and the relationship between apoptosis and the pathogenesis of diabetic nephropathy. Methods High glucose was induced in rats by intravenous glucose infusion. The apoptosis of rat renal cortical cells and the expression of related gene proteins were detected by agarose gel electrophoresis and flow cytometry. Results After a large amount of intravenous infusion of glucose, the apoptosis of renal cortical cells in hyperglycemic rats was increased, the expression of bcl-2 gene was down-regulated and the expression of bax and Fas genes was up-regulated. However, there was no significant difference between the two groups. Conclusion High osmotic pressure may be one of the mechanisms of apoptosis induced by hyperglycemia. Apoptosis is closely related to the occurrence of early diabetic nephropathy
