用离子选择电极通用分沂方法研究了IgG与HRP标记的抗IgG间的免疫沉淀反应。由于HRP可以催化H_2O_2氧化KI的反应,致使溶液中I~-离子活度降低,因此用碘离子选择电极监测I~-离子活度降低以测定IgG与HRP—抗IgG结合形成的沉淀物中HRP的活性,就可以间接求得沉淀的量。本文得到与免疫沉淀曲线相符合的工作曲线,为定量测定IgG抗原提供了依据。文中还对酶免疫电化学传感器进行了讨论。
The immunoprecipitation reaction between IgG and HRP-labeled anti-IgG was studied using ion-selective electrode universal method. Because HRP can catalyze the reaction of H 2 O 2 to oxidize KI, resulting in a decrease in the activity of I- ions in the solution, an Iodine-selective electrode was used to monitor the decrease in I- ion activity to determine HRP in precipitates formed by the combination of IgG and HRP-anti-IgG. The activity can be obtained indirectly from the amount of precipitation. In this paper, a working curve that accords with the immunoprecipitation curve is obtained, which provides a basis for the quantitative determination of IgG antigens. Enzyme immunoelectrochemical sensors are also discussed in this paper.
